The DAI and histological scores of mice treated with probiotics were much better than those did not treat, and the effect of mucosa protection was better in L.acidophilus treated mice.

Abstract

Objective To establish DSS-induced acute ulcerative colitis(UC) mouse models and then to study the effect of B.adolescentis and L.acidophilus on UC mouse models. Methods Altogether 50 specific pathogen free,20 ± 2 g,6-8 week-old BALB/c female mice were randomly allocated into 5 groups: normal control group(N-7),L.acidophilus-treated group(L-7),B.adolescentis-treated group(B-7),sulfasalazine(salazosulfapyridine,SASP)-treated group(S-7) and NS control group(NS-7).With the exception of normal control mice,40 BABL/c mice were induced by 2.5% DSS water for 7 days free drinking. These mice were given with 0.2ml B.adolescentis/L.acidophilus/SASP/NS via gavage once a day for 7 days, while normal controls let untreated. These animals were recorded daily both in weight, fecal trait and occult blood(o-toluidine),and then disease active index(DAI) was accounted.Animals were killed on 8th day morning.Whole colon(from colo-cecal junction to anus) was isolated for examination of length and pieces for hematoxylin and eosin staining, and histological scores were measured. Results B.adolescentis and L.acidophilus could relieve pathogenetic condition of DSS-induced mice. The DAI and histological scores of mice treated with probiotics were much better than those did not treat. The effect of mucosa protection was better in L.acidophilus treated mice. Conclusion The B.adolescentis and L.acidophilus can reduce the inflammation of active UC,and L.acidophilus has more benefits in inducing the relief of active UC.
