Abstract

In the RNA polymerase reaction templated by rat liver or Walker tumor chromatin, addition of a non-histone protein fraction that is homologous to the tissue of chromatin stimulates RNA synthesis in vitro to a greater extent than the heterologous non-histone protein fraction. DNA · RNA hybridization experiments of the transcribed RNA show (1) that RNA synthesized from activated chromatin is transcribed from additional DNA sequences; and (2) that the homologous non-histone protein fraction activates the transcription of different DNA sequences from those activated by the heterologous non-histone protein fraction. It is concluded that such non-histone protein fractions are capable of differentially augmenting transcription in vitro from DNA sequences of chromatin.
