Abstract
Summary and Conclusions The effect of combinations of chemical compounds in cancer chemotherapy has been investigated. By the use of the guanine analog, 5-amino-7-hydroxy-1 H - v -triazolo ( d ) pyrimidine, together with either desoxypyridoxine, pteroylglutamic acid, 7-methyl pteroylglutamic acid, or vitamin B 12 , it has been shown that the growth of a transplantable carcinoma of the breast in mice can be inhibited to a greater extent by a combination than by any one of the drugs alone. Evidence has been presented to show that the combinations are not acting by causing host toxicity, for the observed effects are associated with minimal to no weight loss. The possible mechanisms of action are discussed. It is recognized that in no instance did a combination eradicate the tumor being treated; however, the possibility of utilizing more effective agents in combination or of increasing the number of active chemical compounds in combination offers an attractive approach to achieve this end.
