Abstract

The pharmacokinetics of a new 5-HT uptake inhibitor, paroxetine, as well as its reduction of 5-HT in blood have been investigated in man. The pharmacokinetic studies revealed a good fit to a one compartment open model with elimination half life around 16 hours and an almost complete metabolism. Studies in patients showed a rather small interpatient variation in steady state plasma concentrations. Maximal reduction of 5-HT in blood was seen after treatment with paroxetine 25 mg orally once a day for around 10 days in volunteers as well as in patients.
