Abstract
A new chemical method which separates and measures quantitatively estriol, 16-epiestriol, estradiol-17β and estrone in male and female non-pregnancy urine is described. This method involves acid hydrolysis, extraction with ether-toluene mixture, separation of phenolic fraction from the neutral fraction, purification of the phenolic fraction by refluxing with alkali and alkali washes, chromatographic separation of estriol, 16-epiestriol, estradiol-17β and estrone using partially esterified Amberlite IRC-50 as the adsorbent and a mixture of ethanol and water (3:2 by vol.) as the eluting solvent. The purified fractions are measured by the fluorometric method.Recoveries of estriol, 16-epiestriol and estradiol-17β(6-4μg) added to normal male urine before acid hydrolysis were, on the average, from 70 to 76% and that of estrone (4μg) was 61%.
