Abstract
HYPERBARIC oxygen is known to be mutagenic towards Escherichia coli1–3. But, can oxygen be mutagenic at tensions physiological to man? We have studied this question using oxygen-sensitive mutants of the histidine auxotroph strain TA100 of Salmonella typhimurium constructed by Ames and his coworkers as part of a test system for carcinogens4. We find that oxygen tensions physiological to higher animals are mutagenic towards these organisms.

It is likely that both O 2 − and H 2 O 2 are important agents of oxygen toxicity, and in accord with this conclusion were the observations that augmented intracellular levels of these enzymes correlated with increased resistance towards oxygen damage, whereas increased respiratory capacity correlation with increased sensitivity towards hyperbaric oxygen.

The results support the hypothesis that high pressures of oxygen are mutagenic in E. coli by supporting the possibility that at the time of pressure increase relatively more mutant than normal cells were in the resistant state and that during the period of exposure these might divide, thus giving an increase in mutant frequency and even a increase in the total number of mutants.
