Abstract
The characteristic effects of fluorine and fluoroalkyl substituents on the physico-chemical properties of molecules that are important to biological activity of fluorinated compounds are highlighted. The influence of fluorination on acidity, hydrogen-bonding, and lipophilicity that affect compound absorption and distribution is described. The current perspectives on fluorine steric interactions and the controversial role of hydrogen-bonding involving the CF bond are discussed.

Evidence is presented that a DNA polymerase can exert high fidelity even when a base pair completely lacks conventional hydrogen bonds, and that nucleoside 1 (also called F) serves as a very good template for DNA synthesis.

This account of uses of fluorine in chemotherapy is the sometimes idiosyncratic view of a practicing medicinal chemist who has no special expertise or commitment to organofluorine chemistry, but who, like many or perhaps most medicinal chemists, has become increasingly keen to investigate fluorine-containing compounds in the search for improved drugs.
