Abstract

Cortisol (F), corticosterone (B), 11-deoxycortisol (S), and cortisone (E) can be measured by the present assay in a single extract of 2.0 ml of plasma. Separation on an LH-20 Sephadex column and quantitation by competitive protein-binding allow individual assay of these steroids without interference from other endogenous steroids, or from dexamethasone and prednisone. F and B concentrations are shown to respond appropriately to changes in pituitary activity. F and S values change markedly in patients after overnight metyrapone ingestion. Cortisone was found in the plasma of normal subjects, responded to changes in pituitary activity, and was rapidly converted to F after oral intake.
