Abstract
This chapter discusses on the evidence from in vivo studies which support the conclusion that nicotine influences the secretion of catecholamines and 5-hydroxytryptamine (5-HT) in the brain. The results of recent experiments are considered to investigate the role of the mesolimbic dopamine (DA) and hippocampal 5-HT systems in the psychopharmacological responses to the drug. The effects of nicotine on brain noradrenaline and brain dopamine systems are discussed. The administration of nicotine is also reported to stimulate DA turnover in rodent brain. DA turnover was increased in both the nucleus accumbens and medial striatum following the intraperitoneal injection of 1 mg/kg nicotine whereas the administration of a lower dose (0.3 mg/kg) exerted a significant effect only in the nucleus accumbens. The effects of acute nicotine on DA turnover suggested that the drug stimulated DA secretion in the brain and that the mesolimbic system was more sensitive to the drug than the nigrostriatal system.
