Abstract
The effects of ionizing radiation on the kinetics of semiconservative DNA replication were studied in synchronous populations of Chinese hamster ovary cells by isopycnic centrifugation of DNA, which was density labeled after radiation insult. The proportion of DNA replicated was determined by measuring the amount of DNA that acquired hybrid buoyant density in CsCl gradients. Cells irradiated during mitosis or early G1 with either 150, 300, or 700 rads exhibited a 0.5-hr, dose-independent delay in the initiation of semiconservative DNA replication. Once initiated, however, the kinetics of DNA replication were unaffected in these populations. As cells moved from early G1 to late G1 ionizing radiation effects on the initiation of DNA replication disappeared. Cells, irradiated in late G1 or S phase with doses up to 700 rads, demonstrated no anomalous behavior, either in the initiation or duration of semiconservative DNA replication.
