Abstract

Es wird vergleichend die Substratoxydation durch den Plexus chorioideus und Hirnrindenschnitte vom Kaninchenin vitro geprüft. Die Bestimmung des Sauerstoffverbrauches erfolgte mit der direkten Warburg-Methode. Folgende Substrate wurden verwendet:d-Glukose,d-Ribose,d-Fruktose,d-Sorbit, Na-Succinat, Na-Pyruvat, Na-dl-Laktat, Na-Glutamat, γ-Aminobuttersäure (GABA),l (-)-Äpfelsäure,l(+)-Valin. Der Qo2 des Plexus liegt bei allen Substraten deutlichüber dem des Cortex. Während Glukose und Fruktose praktisch gleichartig metabolisiert werden, wird Sorbit vom Pl. chor. ebenso wie Glukose oxydiert, von der Hirnrinde jedoch wesentlich geringer. Ribose,l(+)-Valin undl(−)-Äpfelsäure zeigen praktisch keinen Einfluss auf den Sauerstoffverbrauch der Gewebepräparate. Pyruvat und Laktat werden durch den Plexus intensiver als Glukose, durch den Cortex jedoch wesentlich geringer als diese oxydiert. Glutamat und GABA metabolisiert sowohl der Cortex als auch der Plexus geringer als Glukose. Überraschend war der Einfluss von Succinat. Während diese Substanz von den Rindenschnitten wesentlich geringer als Glukose oxydiert wird, schnellt die Atmung des Plexus auf das Fünffache der Kontrollen unter Glukose empor. Der Qo2 des Pl. chor. bei einer Succinatkonzentration von 10 mM liegt mit 48,6±2,6 μl O2/mg TG × hüber den höchsten bislang bekannten Werten etwa für die Retina oder den embryonalen Herzmuskel. The oxygen consumption of choroid plexus and brain cortex slices of the rabbit was comparatively studied in the presence of various substrates by measuring the respiration of the tissue with the conventional Warburg manometric apparatus. The following substrates are used:d-glucose;d-ribose,d-sorbitol; the sodium salts of succinate, pyruvate, lactate, glutamate; γ-aminobutyric acid; malic acid; valine. The rate of oxygen consumption by the choroid plexus exceeds that by the brain cortex significantly. The rates with glucose and fructose are identical; ribose, malic acid and valine exercise little or no effect in the rate of oxygen consumption. Pyruvate and lactate yield a much higher rate of the choroid plexus, whereas the respiration of brain cortex slices was less than that found in the presence of glucose. The oxygen uptake by both tissues in the presence of GABA or glutamate was less than that in the presence of glucose. Succinate increases the respiration of the choroid plexus to a very surprising extent; the rate of oxygen consumption reaches the high value of 48,6±2,6 μl O2/mg dry weight × h. The oxygen uptake by cortex slices was not at all affected in the presence of succinate.
